About this study

The researchers are doing this study to find out whether the combination of LBP-MARCO (Microbiota Augmentation to Reestablish Commensal Organisms) and ruxolitinib is a safe treatment for people with graft-versus-host disease (GVHD) after receiving a stem cell (allogeneic hematopoietic cell transplantation).

What participants receive

What participants receive. In a randomised study each person is assigned to one group by chance.

  • RuxolitinibDrug

    Also called Jakafi®

Ruxolitinib will be initiated concurrently with LBP treatment at a starting dose of 10 mg twice daily and will continue for at least 28 days.

  • LBP- MARCOOther

The LBP-MARCO consists of five capsules of individual bacterial strains that will be given at least 1 day apart from one another and within 7-10 days of the first dose.

Study groups in NCT07913672
Study groupRoleWhat they receive
Consortia AExperimentalParticipants in Cohort 1 will receive LBP-MARCO with the same combination of 7 specific types of bacteria.
Receives: Drug: Ruxolitinib; Other: LBP- MARCO
Consortia BExperimentalIf the bacteria do not grow or produce enough metabolites, the next group of participants (Cohort 2) will get LBP-MARCO with a different combination of bacteria.
Receives: Drug: Ruxolitinib; Other: LBP- MARCO
Consortia CExperimentalIf that combination does not grow in participants' guts, the next group of participants (Cohort 3) will get LBP-MARCO with a different combination of the bacteria.
Receives: Drug: Ruxolitinib; Other: LBP- MARCO

What is being measured

The primary outcome is the main question the study is designed to answer.

  1. incidence and severity of SAEs

Grade 2-4 infections by CTCAE v6 either clinically or microbiologically will be assessed throughout the study until day +100. Causality and relationship with aGVHD, LBP-MARCO, and ruxolitinib will be determined.

    Measured: up to 100 days

Secondary outcomes (1)
  1. Microbiota engraftment/PK

is defined as the detection of > or = to 5 of the 7 consortia commensal strains including butyrate producing taxa detected in stool samples

    Measured: day +7 and/or +14

Questions about this trial

What is this clinical trial studying?

A Phase 1 study testing Ruxolitinib and LBP- MARCO in people with Transplant Failure or Rejection of Bone Marrow (Graft vs Host Disease, GVHD), run by Memorial Sloan Kettering Cancer Center at 7 sites in United States.

The full registered description is in the About this study section and the official record.

Who may be eligible to take part?

The sponsor lists 12 inclusion and 12 exclusion criteria. Eligibility is decided by the study team after screening, so read the full criteria and contact a listed site.

Is this clinical trial currently recruiting?

The record says: looking for participants now, as of the sponsor's last update on 3 Nov 2026. Status can change between updates, so confirm with the study contact before making plans.

Where is the study taking place?

The record lists 7 sites in United States. Whether a site is taking part right now should be confirmed with that site. See every site.

What are the possible benefits and risks?

They depend on the study and the treatment, and no study can promise a benefit. The study team explains the known risks during informed consent, before anyone joins; the record itself does not list them.

What does this phase mean?

Phase 1 studies test a treatment in a small group of people for the first time or in a new way, mainly to check safety and find the right dose.

Not every study follows the same path, and some studies (observational studies, device studies) have no phase.

Are results available for this study?

This study is still under way. Results are not available in the source record yet.

How can a doctor refer a patient or contact the research team?

Contact Doris Ponce, MD (646-608-3739, ponced@mskcc.org), or a site near the patient. AzTrials does not enrol participants or take referrals.

What does the primary outcome measure?

incidence and severity of SAEs (time frame: up to 100 days). It is what the study was designed to measure, not a finding.

Where can I read the original study information?
Does a listing mean the treatment is approved or proven to work?

No. A listing means the study is registered. It does not show that the treatment is safe, works, or is approved by a regulator.

How often is this information updated?

AzTrials copies changed records from ClinicalTrials.gov every night and reloads the whole registry every week. The sponsor last updated this record on 3 Nov 2026.
