About this study

Official title: A Phase 3 Randomized, Double-blind, Placebo-controlled Study of JNJ-101556143, a Regulated Induced Proximity Targeting Chimera (RIPTAC™) Therapeutic Plus Best Supportive Care Versus Best Supportive Care in Participants With Metastatic Androgen Pathway Modulation-Resistant Prostate Cancer

The purpose of this study is to evaluate how well JNJ-101556143 plus Best Supportive Care (BSC) improves overall survival (the length of time a participant remains alive) compared with placebo plus BSC in participants with metastatic androgen pathway modulation-resistant prostate cancer (mAPMR). mAPMR is an advanced form of prostate cancer that has spread to other parts of the body and no longer responds to hormone treatments designed to block male hormones (androgens).

What is being measured

The primary outcome is the main question the study is designed to answer.

  1. Overall Survival (OS)

OS is defined as the time from randomization to date of death from any cause.

    Measured: Up to 21 months

Secondary outcomes (8)
  1. Radiographic Progression-Free Survival (rPFS)

rPFS is defined as the time from the date of randomization until the date of radiographic disease progression or death, whichever comes first as per response evaluation criteria in solid tumors version 1.1 (RECIST v1.1) and Prostate Cancer Working Group 4 (PCWG4) criteria.

    Measured: Up to 21 months

  2. Progression-Free Survival (PFS)

PFS is defined as the time from randomization until disease progression or death, whichever occurs first.

    Measured: Up to 21 months

  3. Time to Symptomatic Progression (TSP)

TSP is defined as the time from the date of randomization to the date of the first occurrence of any of the following: The use of external beam radiation for palliative treatment; need for tumor-related orthopedic surgical intervention; other cancer-related procedures; cancer-related morbid events and initiation of a new systemic anticancer therapy because of cancer symptoms.

    Measured: Up to 21 months

  4. Time to Skeletal-Related Events (TSRE)

TSRE is defined as the time from the date of randomization to the date of the first occurrence of any of the following (whichever occurs first):

    • The use of external beam radiation therapy to relieve skeletal symptoms.
    • The need for tumor-related orthopedic surgical intervention.
    • The occurrence of new bone fractures (cancer-related; vertebral or non-vertebral).
    • The occurrence of tumor-related spinal cord compression.

    Measured: Up to 21 months

  5. Time to Pain Progression (TTPP)

TTPP is defined as the time from the date of randomization to first observation of pain progression. Pain progression is defined as an increase of >=2 points from baseline in the BPI-SF worst pain intensity (item 3) observed at 2 consecutive evaluations >=3 weeks apart with no decrease in analgesic dose.

    Measured: Up to 21 months

  6. Number of Participants with Treatment-Emergent Adverse Event (TEAE) by Severity

An adverse event (AE) is any untoward medical occurrence in a participant who received study drug without regard to possibility of causal relationship. An SAE is an AE resulting in any of the following outcomes or deemed significant for any other reason: death; initial or prolonged inpatient hospitalization; life-threatening experience (immediate risk of dying); persistent or significant disability/incapacity; congenital anomaly. Severity will be graded according to the National Cancer Institute Common Terminology Criteria for Adverse Events (NCI-CTCAE). Severity scale defined as Grade 1= Mild, Grade 2= Moderate, Grade 3= Severe, Grade 4= Life-threatening and Grade 5= Death related to adverse event.

    Measured: Up to 21 months

  7. Number of Participants with Abnormalities in Clinical Laboratory Tests

Participants with abnormalities in clinical laboratory tests will be reported.

    Measured: Up to 21 months

  8. Plasma Concentration of JNJ-101556143

Plasma samples will be analyzed to determine concentrations of JNJ-101556143 using a validated LC-MS/MS method.

Measured: Pre-dose for Cycles 1 to 4; 2 to 3 hours and 4 to 8 hours Post-dose from Cycles 1 to 2 (each cycle duration is 28 days)

Questions about this trial

What is this clinical trial studying?

A Phase 3 study testing JNJ-101556143 in people with Prostatic Neoplasms, run by Janssen Research & Development, LLC at 4 sites in 2 countries.

The full registered description is in the About this study section and the official record.

Who may be eligible to take part?

The sponsor lists 6 inclusion and 5 exclusion criteria. Eligibility is decided by the study team after screening, so read the full criteria and contact a listed site.

Is this clinical trial currently recruiting?

The record says: looking for participants now, as of the sponsor's last update on 3 Nov 2026. Status can change between updates, so confirm with the study contact before making plans.

Where is the study taking place?

The record lists 4 sites in United States, Israel. Whether a site is taking part right now should be confirmed with that site. See every site.

What are the possible benefits and risks?

They depend on the study and the treatment, and no study can promise a benefit. Some participants in this study may receive a placebo. The study team explains the known risks during informed consent, before anyone joins; the record itself does not list them.

What does this phase mean?

Phase 3 studies compare the treatment with the current standard treatment or a placebo in a large group, to confirm how well it works and watch for side effects.

Not every study follows the same path, and some studies (observational studies, device studies) have no phase.

Are results available for this study?

This study is still under way. Results are not available in the source record yet.

How can a doctor refer a patient or contact the research team?

Contact Study Contact (844-434-4210, Participate-In-This-Study1@its.jnj.com), or a site near the patient. AzTrials does not enrol participants or take referrals.

What does the primary outcome measure?

Overall Survival (OS) (time frame: Up to 21 months). It is what the study was designed to measure, not a finding.

Where can I read the original study information?
Does a listing mean the treatment is approved or proven to work?

No. A listing means the study is registered. It does not show that the treatment is safe, works, or is approved by a regulator.

How often is this information updated?

AzTrials copies changed records from ClinicalTrials.gov every night and reloads the whole registry every week. The sponsor last updated this record on 3 Nov 2026.
