2D:4D Ratio vs Semen Parameters
An observational study of Male, run by Suleyman Demirel University at 1 site in Turkey (Türkiye).
Trial summary
The aim of this study evaluate the relationship between sperm count and the second-to-fourth digit ratio (2D:4D) in male patients. Additionally, the associations between body mass index, hormonal profile, hand morphometric measurements, and semen parameters were investigated. Study hypotheses: H1. Read more about this study

About this study
Official title: Relationship Between 2D:4D Finger Ratio and Semen Parameters in Men
The aim of this study evaluate the relationship between sperm count and the second-to-fourth digit ratio (2D:4D) in male patients. Additionally, the associations between body mass index, hormonal profile, hand morphometric measurements, and semen parameters were investigated. Study hypotheses:
H1. There is a significant relationship between the second-to-fourth digit ratio (2D:4D) and sperm count in male patients.

What participants receive
What participants receive. In a randomised study each person is assigned to one group by chance.
Group 1Other
The study included male individuals aged 18 years and older. Individuals with a history of index or ring finger fractures, finger injuries, arthritis, gout, or rheumatic diseases affecting the hands or fingers, left-handedness, and lack of semen analysis were excluded. All assessments for this group will be conducted by the doctor.Participation rates and measurements were recorded regularly. For each participant,For each participant, BMI, semen parameters, hand measurements, and 2D:4D ratios were evaluated. Hormonal data were also assessed.
| Study group | Role | What they receive |
|---|---|---|
| Male individuals who applied to the Department of Obstetrics and Gynecology | One-time measurements were planned. For each male participant, measurements of the demographics, Body Mass Index (BMI), semen parameters, hand measurements, 2D:4D ratios and hormonal data. Receives: Other: Group 1 |
What is being measured
The primary outcome is the main question the study is designed to answer.
Hand and finger lengths (2d:4d)
Measurements were taken using standardized digital palm photographs taken with the hand placed on a metric surface at a height of 50 cm. The images were analyzed using ImageJ v1.50 software according to international anthropometric protocols (ISAK, WHO, NASA, and the Anthropometric Standardization Reference Guide). Hand length (HL - distance from the wrist crease at the radiocarpal joint to the distal tip of the third finger), hand width (HW - maximum distance between the second and fifth metacarpal heads), wrist width (WW - distance between the radial and ulnar styloid processes), 2D (distance from the metacarpophalangeal crease of the index finger to the fingertip), and 4D (distance from the metacarpophalangeal crease of the ring finger to the fingertip) were measured. The 2D:4D ratio was calculated by dividing the length of the second finger by the length of the fourth finger.
Measured: Baseline (Day 0)
Semen analysis
Semen samples were collected by masturbation after 2-7 days of sexual abstinence using sterile containers and allowed to liquefy for 30 minutes at 37°C. Ejaculate volume (EjVol) was measured in mL by direct volume assessment and classified as hypospermia (<1.5 mL), normo-volume (1.5-5 mL), or hyperspermia (>5 mL). Sperm concentration (SC) was calculated as total sperm count/semen volume and categorized as severe oligozoospermia (<5 million/mL), mild-moderate oligozoospermia (5-15 million/mL), normozoospermia (15-200 million/mL), or polyspermia (>200 million/mL).Total motility (ABC) was defined as the percentage of progressive (A+B) and non-progressive (C) motile sperm and classified as severe asthenozoospermia (<20%), moderate motility impairment (20-39%), borderline normal motility (40-60%), and good motility (>60%). Progressive motility (PM) was evaluated as severe (<10%), reduced (10-31%), normal (32-50%), or good (>50%).
Measured: Baseline (Day 0)
Hormone analysis
Hormone levels were measured by Enzyme-Linked Immunosorbent Assay (ELISA) from fasting venous serum samples, and the relationships between hand/finger lengths and ratios were evaluated.Hormonal evaluation included the examination of folic acid (FA), follicle-stimulating hormone (FSH), luteinizing hormone (LH), prolactin (PRL), and total testosterone (TT) levels.
Measured: Baseline (Day 0)

They depend on the study and the treatment, and no study can promise a benefit. The study team explains the known risks during informed consent, before anyone joins; the record itself does not list them.
Are results available for this study?
The study is completed, but results are not available in the source record. That does not mean the treatment did or did not work.
How can a doctor refer a patient or contact the research team?
The record lists no contact. The official record names the sponsor, who can be asked directly. AzTrials does not enrol participants or take referrals.
What does the primary outcome measure?
Hand and finger lengths (2d:4d) (time frame: Baseline (Day 0)). The study lists 3 primary outcomes in all. It is what the study was designed to measure, not a finding.
Where can I read the original study information?
Does a listing mean the treatment is approved or proven to work?
No. A listing means the study is registered. It does not show that the treatment is safe, works, or is approved by a regulator.
How often is this information updated?
AzTrials copies changed records from ClinicalTrials.gov every night and reloads the whole registry every week. The sponsor last updated this record on 26 Oct 2026.
