About this study
Official title: A Phase 1b/2, Open-Label, Multicenter, Single-Arm Study of Ivonescimab in Combination With Pemetrexed and Platinum-Based Chemotherapy as First-Line Treatment in Patients With Advanced Malignant Pleural Mesothelioma
This Phase 1b/2, open-label, multicenter, single-arm study evaluates the safety and efficacy of ivonescimab in combination with pemetrexed and platinum-based chemotherapy as first-line treatment for advanced malignant pleural mesothelioma. Phase 1b evaluates the candidate recommended Phase 2 dose (RP2D), followed by Phase 2 evaluation at the confirmed RP2D.
Read the detailed description
In Phase 1b, ivonescimab 20 mg/kg, selected as the candidate RP2D based on prior Phase 1a results, is administered every 3 weeks in combination with pemetrexed 500 mg/m² and cisplatin 75 mg/m² or carboplatin AUC 5 for 4 to 6 cycles. Phase 1b further evaluates safety and preliminary efficacy and confirms the RP2D. In Phase 2, participants receive ivonescimab at the confirmed RP2D with the same chemotherapy regimen, followed by maintenance treatment with ivonescimab with or without pemetrexed every 3 weeks. Objective response rate (ORR) is the primary efficacy endpoint of Phase 2. Peripheral blood, pleural effusion, and pleural tissue samples will be collected for exploratory analyses.

What participants receive
What participants receive. In a randomised study each person is assigned to one group by chance.
Ivonescimab Combined With ChemotherapyDrug
In Phase 1b, ivonescimab 20 mg/kg is administered intravenously every 3 weeks (Q3W) in combination with pemetrexed 500 mg/m² and cisplatin 75 mg/m² or carboplatin AUC 5 for 4 to 6 cycles. In Phase 2, ivonescimab is administered at the confirmed RP2D with the same chemotherapy regimen, followed by maintenance treatment with ivonescimab with or without pemetrexed Q3W.
| Study group | Role | What they receive |
|---|---|---|
| Ivonescimab Plus Pemetrexed and Platinum-Based Chemotherapy | Experimental | A single experimental arm consisting of a Phase 1b fixed-dose expansion followed by a Phase 2 efficacy evaluation. Receives: Drug: Ivonescimab Combined With Chemotherapy |

Questions about this trial
What is this clinical trial studying?
A Phase 1/2 study testing Ivonescimab Combined With Chemotherapy in people with Malignant Pleural Mesothelioma, Advanced, run by Ruijin Hospital at 3 sites in China.
The full registered description is in the About this study section and the official record.
Who may be eligible to take part?
The sponsor lists 8 inclusion and 7 exclusion criteria. Eligibility is decided by the study team after screening, so read the full criteria and contact a listed site.
Is this clinical trial currently recruiting?
The record says: looking for participants now, as of the sponsor's last update on 2 Nov 2026. Status can change between updates, so confirm with the study contact before making plans.
Where is the study taking place?
The record lists 3 sites in China. Whether a site is taking part right now should be confirmed with that site. See every site.
What are the possible benefits and risks?
They depend on the study and the treatment, and no study can promise a benefit. The study team explains the known risks during informed consent, before anyone joins; the record itself does not list them.
What does this combined phase mean?
Phase 1 studies test a treatment in a small group of people for the first time or in a new way, mainly to check safety and find the right dose.
Phase 2 studies give the treatment to a larger group to see whether it has the intended effect and to keep checking safety.
Not every study follows the same path, and some studies (observational studies, device studies) have no phase.
Are results available for this study?
This study is still under way. Results are not available in the source record yet.
How can a doctor refer a patient or contact the research team?
Contact Yi Xiang, MD, PhD (+86-21-64370045, xiangyiht@163.com), or a site near the patient. AzTrials does not enrol participants or take referrals.
What does the primary outcome measure?
Dose-Limiting Toxicities in Phase 1b (time frame: From the first dose through Day 28.). The study lists 2 primary outcomes in all. It is what the study was designed to measure, not a finding.
Where can I read the original study information?
Does a listing mean the treatment is approved or proven to work?
No. A listing means the study is registered. It does not show that the treatment is safe, works, or is approved by a regulator.
How often is this information updated?
AzTrials copies changed records from ClinicalTrials.gov every night and reloads the whole registry every week. The sponsor last updated this record on 2 Nov 2026.
