About this study

Official title: A Phase 4 Open-label, Single-arm Study to Evaluate the Safety and Immunogenicity of an Omicron Subvariant-based SARS-CoV-2 Recombinant Spike (rS) Vaccine Adjuvanted With Matrix-M in Participants ≥ 65 Years of Age and Participants 12 Through 64 Years of Age Who Are at Increased Risk for Severe COVID-19 Illness

The purpose of this study is to evaluate the safety and immunogenicity of a single dose of Sanofi's Omicron subvariant-based Nuvaxovid in participants ≥ 65 years of age and participants 12 through 64 years of age who have at least one underlying condition that puts them at increased risk for severe outcomes of COVID-19 and who have been previously vaccinated with a COVID-19 vaccine ≥ 90 days prior to study vaccination.

Study details include:

  • The study duration will be approximately 3 months per participant.
  • Study intervention will be administered via a single intramuscular (IM) injection into the deltoid muscle on Day 1.
  • The visit frequency will be Day 1, Day 9 telephone call, Day 29, and Day 91 telephone call (3-month safety follow-up).

What is being measured

The primary outcome is the main question the study is designed to answer.

  1. Geometric Mean Fold Rise (GMFR) in pseudovirus neutralizing antibody titers (ID50) against the Omicron subvariant included in the vaccine formulation from pre-vaccination to 28 days post-vaccination.

Superior neutralizing antibody response is defined as a GMFR with a lower bound of the two-sided 95% confidence interval greater than 4.0 within each age cohort.

Measured: Day 1 (pre-vaccination) and Day 29 (28 days after vaccination)

Secondary outcomes (11)
  1. Geometric Mean Titers (GMTs) of Pseudovirus Neutralizing Antibodies Against the Omicron Subvariant Included in the Vaccine Formulation

Pseudovirus neutralizing antibody responses against the Omicron subvariant included in the vaccine formulation will be measured as geometric mean titers (GMTs).

Measured: Day 1 (pre-vaccination) and Day 29 (28 days after vaccination)

  2. Seroresponse Rate (SRR) in Pseudovirus Neutralizing Antibody Titers Against the Omicron Subvariant Used in the Vaccine Formulation

Seroresponse is defined as a pre-vaccination antibody titer below the lower limit of quantitation (LLOQ) and a post-vaccination antibody titer ≥ 4 × LLOQ; at least a 4-fold rise when the pre-vaccination titer is ≥ LLOQ and < 4 × LLOQ; or at least a 2-fold rise when the pre-vaccination titer is ≥ 4 × LLOQ.

Measured: Day 29 (28 days after vaccination)

  3. Presence of Immediate Adverse Events (AEs)

Number of participants reporting immediate adverse events within 30 minutes following study vaccination.

Measured: Within 30 minutes after vaccination on Day 1

  4. Presence of Solicited Injection Site Reactions

Number of participants reporting solicited injection site reactions during the 8-day period including the day of vaccination.

Measured: Day 1 through Day 8

  5. Presence of Solicited Systemic Reactions

Number of participants reporting solicited systemic reactions during the 8-day period including the day of vaccination.

Measured: Day 1 through Day 8

  6. Presence of Unsolicited Adverse Events (AEs)

Number of participants reporting unsolicited adverse events during the 28-day period including the day of vaccination.

Measured: Day 1 through Day 28

  7. Presence of Medically Attended Adverse Events (MAAEs)

Number of participants reporting medically attended adverse events (MAAEs) during the 28-day period including the day of vaccination.

Measured: Day 1 through Day 28

  8. Presence of Related Medically Attended Adverse Events (MAAEs)

Number of participants reporting medically attended adverse events considered related to the study intervention from Day 29 through the end of the study.

Measured: Day 29 through approximately Day 91

  9. Presence of Adverse Events of Special Interest (AESIs)

Number of participants reporting adverse events of special interest (AESIs) throughout the study. Protocol-defined AESIs include anaphylaxis; myocarditis, pericarditis, or myopericarditis; and new onset or worsening of potential immune-mediated disease.

Measured: Day 1 through approximately Day 91

  10. Presence of Serious Adverse Events (SAEs)

Number of participants reporting serious adverse events (SAEs) throughout the study.

Measured: Day 1 through approximately Day 91

  11. Presence of Adverse Events (AEs) Leading to Study Discontinuation

Number of participants reporting adverse events leading to study discontinuation throughout the study.

Measured: Day 1 through approximately Day 91

Questions about this trial

What is this clinical trial studying?

A Phase 4 study testing Omicron Subvariant-Based SARS-CoV-2 Recombinant Spike Vaccine Adjuvanted With Matrix-M in people with COVID-19, run by Sanofi at 20 sites in United States.

The full registered description is in the About this study section and the official record.

Who may be eligible to take part?

The sponsor lists 4 inclusion and 8 exclusion criteria. Eligibility is decided by the study team after screening, so read the full criteria and contact a listed site.

Is this clinical trial currently recruiting?

The record says: looking for participants now, as of the sponsor's last update on 15 Oct 2026. Status can change between updates, so confirm with the study contact before making plans.

Where is the study taking place?

The record lists 20 sites in United States. Whether a site is taking part right now should be confirmed with that site. See every site.

What are the possible benefits and risks?

They depend on the study and the treatment, and no study can promise a benefit. The study team explains the known risks during informed consent, before anyone joins; the record itself does not list them.

What does this phase mean?

Phase 4 studies take place after a treatment is approved, to learn more about its long-term benefits and risks in everyday use.

Not every study follows the same path, and some studies (observational studies, device studies) have no phase.

Are results available for this study?

This study is still under way. Results are not available in the source record yet.

How can a doctor refer a patient or contact the research team?

Contact Trial Transparency email recommended (Toll free number for US & Canada) (800-633-1610 ext. 6, contact-us@sanofi.com), or a site near the patient. AzTrials does not enrol participants or take referrals.

What does the primary outcome measure?

Geometric Mean Fold Rise (GMFR) in pseudovirus neutralizing antibody titers (ID50) against the Omicron subvariant included in the vaccine formulation from pre-vaccination to 28 days post-vaccination. (time frame: Day 1 (pre-vaccination) and Day 29 (28 days after vaccination)). It is what the study was designed to measure, not a finding.

Where can I read the original study information?
Does a listing mean the treatment is approved or proven to work?

No. A listing means the study is registered. It does not show that the treatment is safe, works, or is approved by a regulator.

How often is this information updated?

AzTrials copies changed records from ClinicalTrials.gov every night and reloads the whole registry every week. The sponsor last updated this record on 15 Oct 2026.
