About this study
Official title: A Phase 1/2a, Multicenter, Open-label Study to Evaluate the Safety, Tolerability, Pharmacokinetics, and Preliminary Anti-tumor Activity of JAB-23E73 Monotherapy and in Combination With Other Agents in Adult Patients With Advanced Solid Tumors With KRAS Alteration
This study is to evaluate the safety and tolerability of pan-KRAS inhibitor JAB-23E73 Monotherapy and in Combination with Other Agents in adult participants with advanced solid tumors
Read the detailed description
This is a multi-part, open-label, multi-center Phase 1/2a study evaluating JAB-23E73 as monotherapy (Part A) and in combination with other agents (Part B1: with cetuximab) in adult participants with advanced solid tumors.
Part A will evaluate the safety, tolerability, pharmacokinetics, pharmacodynamics, and preliminary anticancer activity of JAB-23E73 as a single agent in adult patients with advanced solid tumors with KRAS alteration. Part A consists of three phases: Phase 1a dose-escalation, Phase 1b dose expansion (dose optimization), and Phase 2a indication expansion.
Part B1 will evaluate the safety, tolerability, PK, PDc, and preliminary anti-tumor efficacy of JAB-23E73 combined with cetuximab in adult participants with locally advanced or metastatic KRAS-altered CRC. Part B1 consists of two phases: Phase 1b dose escalation phase and Phase 2a dose expansion phase.

What is being measured
The primary outcome is the main question the study is designed to answer.
Part A: Number and proportion of patients who experience at least 1 dose-limiting toxicity (DLT) with treatment of JAB-23E73 monotherapy.
A DLT is defined as any toxicities that meet the criteria for a DLT defined in the protocol
Measured: Up to 1 year
Part A: Safety and tolerability of JAB-23E73 monotherapy
Incidence, and severity of adverse events and changes in clinical laboratory tests and examinations.
Measured: Up to 2 years
Part A: Objective response rate (ORR) of JAB-23E73 monotherapy.
ORR, defined as the percentage of patients with the best overall response of confirmed partial response (PR), or complete response (CR) per RECIST v1.1.
Measured: Up to 3 years
Part B1: Safety, tolerability and RP2D of JAB-23E73 in combination with cetuximab.
Incidence, and severity of adverse events and changes in clinical laboratory tests and examinations. RP2D will be determined based on comprehensive available data
Measured: Up to 2 years
Part B1: Objective response rate (ORR) of JAB-23E73 in combination with cetuximab.
ORR, defined as the percentage of patients with the best overall response of confirmed PR, or CR per RECIST v1.1.
Measured: Up to 3 years
Secondary outcomes (4)
Part A: DOR, TTR, PFS, and DCR assessed by investigators, and OS of JAB-23E73 monotherapy
Assessed according to Response Evaluation Criteria in Solid Tumors (RECIST) v1.1.
Measured: Up to 3 years
Part A: Pharmacokinetic (PK) profile of JAB-23E73 monotherapy
Maximum Observed Concentration (Cmax), time to reach maximum concentration (Tmax), and area under the plasma concentration-time curve (AUC), etc
Measured: Up to 2 years
Part B1: DOR, TTR, PFS, and DCR assessed by investigators, and OS of JAB-23E73 in combination with cetuximab
Assessed according to Response Evaluation Criteria in Solid Tumors (RECIST) v1.1.
Measured: Up to 3 years
Part B1: PK profile of JAB-23E73 in combination with cetuximab.
The plasma concentration and PK parameters of JAB-23E73, including but not limited to: Cmax, AUC, and Ctrough.
Measured: Up to 2 years

Questions about this trial
What is this clinical trial studying?
A Phase 1/2 study testing JAB-23E73 and Cetuximab in people with Advanced Solid Tumors, run by Jacobio Pharmaceuticals Co., Ltd. at 4 sites in United States.
The full registered description is in the About this study section and the official record.
Who may be eligible to take part?
The sponsor lists 9 inclusion and 8 exclusion criteria. Eligibility is decided by the study team after screening, so read the full criteria and contact a listed site.
Is this clinical trial currently recruiting?
The record says: looking for participants now, as of the sponsor's last update on 26 Oct 2026. Status can change between updates, so confirm with the study contact before making plans.
Where is the study taking place?
The record lists 4 sites in United States. Whether a site is taking part right now should be confirmed with that site. See every site.
What are the possible benefits and risks?
They depend on the study and the treatment, and no study can promise a benefit. The study team explains the known risks during informed consent, before anyone joins; the record itself does not list them.
What does this combined phase mean?
Phase 1 studies test a treatment in a small group of people for the first time or in a new way, mainly to check safety and find the right dose.
Phase 2 studies give the treatment to a larger group to see whether it has the intended effect and to keep checking safety.
Not every study follows the same path, and some studies (observational studies, device studies) have no phase.
Are results available for this study?
This study is still under way. Results are not available in the source record yet.
How can a doctor refer a patient or contact the research team?
Contact Jacobio Pharmaceuticals ((781) 918-6670, clinicaltrials@jacobiopharma.com), or a site near the patient. AzTrials does not enrol participants or take referrals.
What does the primary outcome measure?
Part A: Number and proportion of patients who experience at least 1 dose-limiting toxicity (DLT) with treatment of JAB-23E73 monotherapy. (time frame: Up to 1 year). The study lists 5 primary outcomes in all. It is what the study was designed to measure, not a finding.
Where can I read the original study information?
Does a listing mean the treatment is approved or proven to work?
No. A listing means the study is registered. It does not show that the treatment is safe, works, or is approved by a regulator.
How often is this information updated?
AzTrials copies changed records from ClinicalTrials.gov every night and reloads the whole registry every week. The sponsor last updated this record on 26 Oct 2026.
